Carreno Busta, the 12th seed, was the first seeded player to progress to the next round after accounting for American qualifier Evan King 6-3 6-2 7-6 (7-5).
Ferrer, the 21st seed, won the first set against Mikhail Kukushkin but the Kazakh qualifier was too good after that point and won 4-6 6-3 6-2 6-1.
South African veteran Kevin Anderson, the 28th seed, beat American qualifier JC Aragone 6-3 6-3 6-1.
Diego Schwartzman, the Argentine 29th seed, overpowered his countryman Carlos Berlocq 6-2 6-1 6-3.
Kyle Edmund of Great Britain caused an upset with a 6-3 7-5 6-3 victory over Dutchman Robin Haase, the 32nd seed.
American Steve Johnson accounted for experienced Spaniard Nicolas Almagro 6-4 7-6 (7-2) 7-6 (7-5) while Frenchman Benoit Paire beat Lukas Lacko of Slovakia 6-3 6-2 7-6 (7-5).
Korean Chung Hyeon defeated Horacio Zeballos of Argentina 3-6 7-6 (10-8) 6-4 6-3 and Florian Mayer of Germany edged Rogerio Dutra Silva of Brazil 7-5 0-6 6-3 6-4.
British qualifier Cameron Norrie was two sets to the good when Russian opponent Dmitry Tursunov retired while Evgeny Donskoy of Russia was also the beneficiary of a retirement.
